Inhibitory effect of Thottea Grandiflora Rottb. on platelet-activating factor receptor binding antagonist activity

Abstract

The extract of Thottea grandiflora RoUb. was evaluated for platelet-activating factor P AF receptor binding antagonist activity using rabbit platelets. The crude methanolic extract of T. grandiflora at a concentration of 200 J..l.g/ml showed strong inhibitory effect on P AF receptor binding antagonist with an inhibition of 95.5 and ICso value of 44 J..l.g/ml. The methanolic extract was fractionated into petroleum ether dichloromethane ethyl acetate and methanol fractions respectively. It was found that the highest inhibitory activity was from the petroleum ether fraction where its inhibition percentage at a concentration of 200 J..l.g/ml was more than 90. The petroleum ether fraction was further fractionated via Vacuum Liquid Chromatography VLC to obtain 10 fractions namely TGI to TGI0. The inhibitory effect of each fraction was evaluated. Fraction TGlO at the concentration of 200 J..l.g/ml showed strong inhibitory effect on P AF receptor binding activity with an inhibition of 99. Fractions TGl TG2 TG8 and TG9 exhibited moderate activities of 76 67 66 and 65 respectively. Fraction TG3 showed low activity with a percentage of 45 while fraction TG4 TG5 TG6 and TG7 indicated negative inhibition.
