Abstract
Centella asiatica known as pegaga in Malaysia has been in use since ancient times especially in the Ayurvedic medical system of India the folk medicine of China and Madagascar. In Malaysia it is commonly consumed as an ulam among the Malays a cooling drink by the Chinese and a brain tonic by the Indians. In this study crude extracts in methanol chloroform n-butanol ethanol and petroleum ether from various accessions of C. asiatica were used to evaluate their anti-proliferative properties on a panel cell lines via the methylene blue bioassay. Using the most active extract mode of action was investigated through TUNEL assay and expressions of pro-apoptosis Bax oncogene and anti-apoptosis Bcl-2 oncogene. The IC50 values of the n-buthanolic petroleum ether and chloroform extracts on ovarian cancer cells CaOV-3 were 43.31 ug/ml 57.75 ug/ml and 66.5 ug/ml respectively. Both n-buthanolic and ethanolic extracts also showed an anti-proliferative effect against epidemoid carcinoma cells KB with IC50 value at 29.5 ug/ml and 64.75 ug/ml respectively. Meanwhile IC50 value of 89.1 ug/ml was recorded by methanolic crude extract against breast cancer cells MCF7. No anti-proliferative effect however was observed with other extracts IC5O _ 99.0 ug/ml. TUNEL Tdt-mediated-nick-end labeling assay Boehringer Mannhaem was used to detect the presence of DNA fragmentation a significant character of apoptosis programmed-cell-death event. The extent of apoptosis was evaluated for the most active extract. The results however were not satisfactory as the system needs to be optimised to reduce background interference.
