Abstract
Henna Lawsonia inermisis an Indian medicinal plant use in traditional medicine for the treatment of various diseases besides its popularity as a natural dye to colour had and hair. Response surface methodology RSMwas performed to optimize the extraction parameters for assessing maximum yield of phenolic antioxidants from henna stems. A first set of single factor experiments allowed for identifying the optimum range of each independent variable affecting total phenolic content TPC extraction efficiency of henna stems namely the solvent type solvent concentration v/v. extraction time min and extraction temperature C. A five level three variable central composite rotatable design CCRD was then applied to investigate the best possible combination effects of acetone concentration extraction time and and extraction temperature on TPC of henna stem extracts. The actual value of the independent variables coded at five levels were selected based on the results of single factor experiments. The determination coefficient R2 value was 0.9648 and its lack of fit was insignificant at 5 level. The optimum extraction conditions were found to be at acetone concentration 47.02 extraction time 47.56 min and extraction temperature 37030C. Under these optimized conditions the experimental maximum yield of TPC was 5232.39mg GAE/100g DW which was in close agreement with predicted values 5231.79mg GAE/100g DW thus indicating the suitability of the models develop and the success of RSM in optimizing the extraction conditions of phenolic antioxidants from henna stems.
