Abstract

The possible activation of the peripheral ATP-sensitive K channels by Mitragyna speciosa Korth methanolic extract MSME was assessed in the rat 1 formalin test. MSME given by intraperitoneal route produced significant P0.05 dose-dependent anti-nociception in the first and the second phase of the test. However local pre-treatment of the paw with glibenclamide an ATP-sensitive K channel inhibitors tetraethylammonium blockers of voltage-dependent K channel and apamin inhibitor of small-conductance Ca2-activated K channel significantly reversed the MSME-induced antinociception. These results suggest that the anti-nociceptive effect of MSME is mediated by the activation of ATP-sensitive K channels in addition to the opening of small-conductance Ca2-activated K channels at peripheral afferent neurons.
