Abstract

Strobilanthes crispa T. Anderson also known locally as pecah beling has been used as a traditional treatment for diabetes. Diabetes is a metabolic disease characterized by hyperglycemia that occurs due to abnormality in insulin secretion insulin action or both. Although many natural supplements are widely used around the world for the treatment of diabetes there are very few scientific reports that support their effectiveness. Thus searching for an effective anti-diabetic agent from natural sources has attracted much interest. This study investigates the insulin-secreting activity of insulinoma cell line treated with S. crispa. Insulinoma cells were cultured at a concentration of 0.2xl06 cells/well in 24-welled plate in the presence and absence of extracts and glybenclamide as positive control at different concentrations 100 10 l ug/ml for 20 minutes. Aliquots were removed and insulin concentrations were estimated by using Insulin Elisa ELISA Kit Biosource. Both petroleum ether and methanol extracts stimulated the secretion of insulin pO.O5 in insulinoma cells at concentrations of 1 10 and 100 ug/ml compared to untreated cells negative control. Aqueous extract of S. crispa increased insulin secretion pO.O5 significantly at concentration of 10 ug/ml. Compared to glybenclamide positive control methanol extract of S. crispa was more effective in promoting secretion of insulin in the treated cells. The results also indicated that at concentration 100 ug/ml methanol extract of S. crispa induced insulin secretion in insulinoma cells up to 22.74. Our preliminary result supported the traditional use of this plant as anti-diabetic agent.
