Sidaguri Sida rhombifolia L. and seledri Apium graveolens L. as anti gout : in vitro in vivo assays and bioactive compounds

Abstract

The ;back to nature slogan; of pharmaceutical industries and community in the world and also the increasing number of patients with gout have supported the effort for discovering the formula of gout treatment using Indonesian medicinal plant. Indonesia has many medicinal plants such as Sidaguri Sida rhombifolia L. and Seledri Apium graveolens L. containing flavonoids and alkaloids which show inhibition of xanthine oxidase enzyme activity while the enzyme itself could convert xanthine compound into uric acid in blood. The research aimed to find formula through assay in vitro and in vivo of Sidaguri and Seledri as anti gout medication; also assay of anti inflammation effect and toxicity of formula. Source material of Sidaguri and Seledri were harvested from five places of different elevations. Sidaguri and Seledri were extracted by water ethanol and extraction to find the crude flavonoids. Results indicated that most extracts could inhibit the activity of xanthine oxidase while the mixed extract could inhibit the xanthine oxidase enzyme higher than single extracts. The best formula is a mixture of flavonoid crude extract of Sidaguri and ethanol extract of Seledri of a certain ratio. The formula could inhibit the activity of xanthine oxidase completely and the inhibition effect was found to be higher than allopurinol or other commercial products. The formula has shown significant effects in decreasing uric acid in rat blood in vivo during a period of one week. The non-toxic formula yielded significant as anti inflammation and phenolic or flavonoid compound as anti gout.
