Abstract
Changes in the cell wall composition of fruit during ripening account for alterations in textural properties which can lead to both desirable and undesirable postharvest qualities. To further understand this mechanism during the ripening period it is important to establish which cell wall polymers are altered significantly as softening develops and determine what types of structural changes are occurring. This paper describes current techniques used in our research laboratory in the analysis of fruit cell walls in relation to ripening of fruit such as papaya paw paw and strawberries. Cell wall materials are isolated after treating frozen fruit powder with phenol/HEPES 4: 1 w/v at pH 6.5. The isolate is then sequentially fractionated using a sequence of solvents with increasing solvation strength. Fractions containing polysaccharides are then studied in detail using gas chromatography. Solid-state l3C cross polarisation magic-angle spinning nuclear magnetic resonance of the isolated fruit cell wall material is also used to explore the three-dimensional arrangement of polysaccharides in the cell walls.
