Abstract

Natural rubber industrial effluent is one of the main environmental concerns especially in Malaysia. Studies conducted on natural rubber industrial effluent shows it contains a valuable protein named hevein. L-cystine is selected to represent hevein in this study as it is a major amino acid in hevein. Aqueous two-phase system ATPS of PEG-salt has been successfully used for separation and purification of protein from various sources such as papaya latex pineapple peel tuna spleen and many more. This study aims to investigate the potential of recovering I-cystine using PEG-ammonium sulphate NH42S04 aqueous system. PEG at molecular weight of 2000 4000 and 6000 were used to investigate the effect of PEG molecular weight on I-cystine partitioning behaviour. It was found that the highest percentage of I-cystine recovery obtained is 78.39 in the bottom phase using 30 wt PEG 6000 - 30 wt NH42S04 aqueous system.
