Abstract
The aim of the present study was to investigate the type of immune response in BALB/c mice that were supplemented with tocotrienol rich fraction from palm oil (TRF) that were either injected (i) with cancer cells or (ii) immunised with a specific antigen like ovalbumin (OVA). Sixty BALB/c mice were divided into three groups of twenty animals each. In each group, the mice were further divided into experimental mice that were supplemented daily with TRF and control, each with ten mice. In group 1, control and experimental mice were injected with 1x102 4T1 mouse mammary cancer cells; while mice in group 2 were immunized s.c. with a single dose of 50µg OVA whilst mice in group 3 were not injected with tumour cells or OVA. In Group 1 mice at the end of 8 weeks, there was a significant delay in the onset, incidence and size of tumours in BALB/c mice supplemented with TRF compared with the controls. There was also a significant increase in IFN-y levels in Group 1 mice that were supplemented with TRF and injected with cancer cells compared to control mice. Experimental mice in Group 2 supplemented with TRF and injected with OVA also produced significant levels of IFN-y. IL-4 production was significantly elevated in TRF supplemented mice compared to control mice however it was eight and four times less than IFN-y in Group 1 and 2 mice respectively. Group 3 animals did not show any differences in IFN-y and IL- 4 levels. These results demonstrate that tocotrienols are able to induce IFN-y and therefore a TH1 response when the mice are subjected to an immune challenge. Further studies will be conducted to evaluate the ability of IFN-y induction as adjuvant therapy.
