Abstract

Cellular activities such as Mitotic Index chromosome counts cell doubling times nuclear DNA content mean cell and nuclear areas were examined to find out whether these activities change during the flowering phase of the plantlets of Petunia in tissue culture system. Our aim was to investigate the relationship between cellular activities and in vitro flowering. From our investigations we found that regeneration and in vitro flowering could only be obtained when leaf and pedicel explants of Petunia hybrida Vilm were used. Leaf explants were cultured on MS supplemented with 1.0 mg/l IAA and 1.0 mg/l zeatin while pedicels from intact plants were cultured on MS supplemented with 1.0 mg/l kinetin in order for flowering to occur. Based on cellular studies it was found that Mitotic Index and chromosome counts increased during flowering as compared to in vivo values. The cell doubling times were shorter no polyploid cells were detected and cell and nuclear sizes were bigger in root meristem cells of flowering plantlets.
