Abstract

Transformation of Melastoma malabathricum with green fluorescent protein GFP and -glucuronidase GUS genes using Agrobacterium-mediated method was studied. The effects of immersion time types of wounding acetosyringone and Agrobacterium concentration known to influence transformation efficiency were assessed. Results were based on the percentage of GFP expression which was observed three days after transformation. Histochemical GUS analysis confirmed that the reporter gene was transferred to the cells and was successfully expressed. In the presence of hygromycin putative transformants containing hygromycinresistant gene survived. The indentation sites of leaves were found to be the major sites of infection. Callus formed at the edges of leaves also showed GFP expression.
