Abstract

Establishment of a highly efficient and widely used tissue culture system for indica rice will accelerate the application of transformation technology. A frequency of obtained callus from seed depends highly on the geno type and stages of seeds. Eight stages of rice seeds variety MR 219 were tested for their callus induction response. By manipulating stages of seed the experiment was conducted to observe their effect on callus response. The stages were: immature/milky half mature mature fresh harvested or 1-6 months after harvest. The callus induction was on the MS basal medium with 30g/L sucrose 3.2g/L phytagel and 1 mg/L 2 4-D in combination with 10 mg/L NAA. Callus responses were obtained for each of the tested stages with a maximum at half mature stage 2.5g callus/plate. The used of seed for callus induction at five months-old and above are not recommended due to the reduction infresh weight of callus.
