Abstract

Callus cultures were initiated from leaf explants of Sentang Azadirachta excelsa Jacob cultured on an induction medium: Murashige and Skoog;s medium supplemented with casein hydrolysate ascrobic acid three amino acids and auxin to cytokinin combinations. Leaf discs spontaneously developed callus and callusing frequency varied between 5-80 depending on the treatments applied. Two types of calli were observed: Callus type1 which was compact nodular pale yellow to creamy white and Callus Type 2 which was Semi-friable pale yellow to yellowish Azadiratwhite callus. Microscopy studies have also revealed morphologically different cellular characteristics of the two calli. Type1 calli were composed of isodiametric thin walled cells and randomly dispersed groups of smaller dividing cells i.e.meristem centers. Conversely Type 2 calli contained smaller sized cells appearing in clusters and each having a prominently larger nucleus.
