Abstract

Seven biological factors affecting gusA gene expression in protocorm-like-body PLB of Dendrobium Sonia were tested. Three-week-old PLB ranging from 3 to 6 mm in size had significantly higher transient GUS activity and PLB of different genotypes exhibited different GUS expression rate. High transient gusA gene expressions were also observed in PLB sub-cultured onto fresh medium prior to transformation with 1.5g DNA. The gusA gene expression was maximal 2 days post-bombardment. Treatment of PLB with sorbitol or mannitol prior bombardment however had no significant effects on gusA gene activity.
