Abstract

Himbacine one of many unique alkaloids isolated from the Australian rainforest tree Galbulimima belgraveana has attracted attention as a lead compound for drug development because of it being a significant muscarinic antagonist with selectivity for M‚‚ receptor sites. Efforts to prepare analogues based on the himbacine structure effective for the treatment of Alzheimer;s disease have so far been unsuccessful but other studies have had success. Also compounds based on the enantiomeric structure of himbacine have shown potent activity as PAR-1 antagonists and may be of use in the treatment of cardiovascular diseases.
