Abstract
Oxalate oxidase OXO which is known to be a true germin has a coding region of 675 base pairs. OXO is involved in a variety of plant processes including germination response to abiotic stress regulation and pathogen infestation. Here we report the isolation of the axa gene from a barley and the use of this gene to construct an OXO over-expression cassette. The axa gene was successfully isolated from BAC clone 450N4 through PCR using specifically designed primers based on the barley OXO cDNA sequence from NCBI. The amplified sequence was identical to the barley axa gene Y14203 through the BLASTp programme and via the BLASTx programme. The OXO sequence encoding 224 amino acids was cloned under the control of CaMV35S promoter in a binary vector pCambia 1305.2. Our local rice Oryza indica MR219 and our own varieties UKMR2 UKMR9 were chosen for the incorporation of the gene via Agrobacterium mediated transformation of embryonic calli. Embryogenic calli were successfully generated on modified MS medium containing 4 mg/L of 2 4-D at 25C.
