Biodegradation of phorbol esters extracted from Jatropha curcas l. kernel by Trichoderma spp.

Abstract

Jatropha curcas L. seeds have the potential as a source of oil for biodiesel production. However the oil as well as the kernel meal residue left after oil extraction contains high levels of phorbol esters which have toxic properties. The kernel meal is a good feed ingredient for livestock as it contains high levels of protein and low levels of fiber but the presence of toxic compounds limit its use. In this study two Trichoderma spp. T. harzianum and T. pseudokoningii were evaluated for their ability to degrade phorbol esters. The kernel fraction was separated from ripe J. curcas seeds and phorbol esters rich fraction was prepared by using methanol extraction. The fungal species were inoculated into 30ml of potato dextrose broth medium containing 84mg of phorbol esters and incubated for 21 days. Growth supernatant was analyzed for phorbol esters at day 0 7 14 and 21 by high performance liquid chromatography using phorbol-12-myristate-13-acetate as the standard. At day 0 the level of phorbol esters was 2.8 mg/ml and the levels decreased gradually with incubation period. At day 21 the amount of phorbol esters decreased by 84 and 81 with T. harzianum and T. pseudokoningii treatment respectively. Based on these findings both fungal species are potential candidates for biological treatment of J. curcas seed kernel.
