Abstract
The present study aims to optimize the carbon and nitrogen requirements for the formulation of a low cost medium for cultivation of a local Ganoderma lucidum Cendawan Senduk in submerged fermentation. The ingredients investigated for cultivation medium were brown sugar and brewery spent yeast. The response surface analysis was used to determine the optimal percentage of carbon and nitrogen in the medium that resulted in the highest mycelia biomass as a response. The model predicted the optimal carbon and nitrogen levels in combination as brown sugar and brewery spent yeast at 10 C and 0.075 N with the mycelial dry weight of 21.50 0.049 g/L. The mycelia biomass values from validation experiments agreed well with the predicted values. Conclusively the response surface analysis using central composite design CCD was determined the optimal medium for G. lucidum cendawan senduk cultivation which composed of 37.25 g/l brewery spent yeast 91.3 g/l brown sugar 0.5 g/l MgSO‚„ 7H20 0.5 g/l KH‚‚PO‚„ and 0.5 g/l K‚‚HPO‚„. The optimum condition of carbon and nitrogen obtained from the statistical design will be useful to develop G. lucidum cendawan senduk cultivation process for efficient production.
