Abstract

An extracellular thermostable protease producer has been successfully isolated from Lojing hotspring Kelantan Malaysia. The isolate was identified as Bacillus subtilis by 16s rRNA analysis. Effect of temperature on the enzyme activity and stability was investigated. The crude protease displayed the maximal activity at 70C. The thermostability profile exhibited that the protease was very stable at 50C and the activity retained 89 of its original activity after heat treatment at 60C for 30 min. The enzyme activity was totally inhibited by phenylmethyl sulphonyl fluoride PMSF suggested that the protease from B. subtilis 50a belongs to the family of serine protease. From the results it showed that B. subtilis 50a is a good extracellular protease producer at high temperature and thus making it as promising thermostable serine protease producer forindustrial use.
