Abstract
Lactococcus lactis is a mesophilic fermentative lactic acid bacterium LAB that has been used extensively in dairy and fermented food industries. This organism also plays a pivotal role as a cell factory for the production of food ingredients neutraceuticals heterologous proteins and vaccine delivery. Proteome of the local isolated Lactococcus lactis M4 strain obtained from the milk of locally-bred cows was studied to elucidate the physiological changes occurring between the growth phases late-exponential and early-stationary of this bacterium when cultivated in an optimum medium. In this study a simple gel-free label-free and advance LCMSE approach was used for qualitative proteomic analysis. Total proteins were extracted and subjected to trypsin digestion. Digested peptides were then separated by LC-nanoESI-MSE alternating low-energy MS and elevated energy MSE. MSE data were processed and the proteins were identified by correlation of tandem mass spectra and Lactococcus protein database available at UniProt using ProteinLynx Global Server PLGS version 2.4. The resulted proteome gives us a better understanding of the protein expression patterns of M4 strain at different growth phases which in turn could provide insight on various applications.
