Abstract
Two molecular technologies the polymerase chains reaction PCR and restriction endonuclease enzyme RE were developed for the management of rhinoceros beetle Orytes rhinoceros using Oryctes Nudivirus OrNV. The PCR is a simple procedure sensitive and accurate in estimating the infection of OrNV on adults and larvae of the beetles. The presence of OrNV was detected from insect gut tissues which contained DNA concentration as low as 2.23 pg l- . The PCR diagnosis showed that infection of OrNV on adults ranged from 30 to 65 while on larvae from 0 to 35. Pre-pupae and pupae are free from the OrNV infection. The RE analysis by the endonuclease enzyme Hindlll identified four types of OrNV named as types A B C and D. Insertion of a single DNA band at size of 15 471 bp has differentiated the type B from other types. The OrNV type B was more pathogenic against the larvae and neonates. In field with palms below 1-year old the introduction of the OrNV type B had successfully reduced the adult population as well as palm damage. The OrNV type B was established as early as 3 months after release MAR and persisted up to 15 MAR. In field with palms more than 3-years old the OrNV infection had gradually increased and maintained at a higher level between 60-90. The adult population reduced and maintained at low levels for a certain period before slowly increasing again to reach the second peak. Virus infection had a weak negative correlation with adult population. Slow reduction on proportion of males was observed possibly due to slow virus transmission and :he adult population may have possibly adapted to the virus infection. The RE analysis showed that the OrNV type B was detected only at 4 MAR. Factors ensuring the success of the virus transmission in the fields were elaborated. Further research work to fully utilise the rhinoceros virus to ensure maximum control of the rhinoceros beetle was also discussed.
