Growth of different Ganoderma isolates on palm extract media and media with various sugar compounds under in-vitro conditions

Abstract

In-vitro growth of different Ganoderma isolates was evaluated on conventional as well as oil palm and coconut extract media. Seven Ganoderma isolates with four G. boninense isolates obtained from oil palm plantations one G. lucidum one G. australe isolated from Schizolobium parahybum Tower Tree and one Ganoderma species from coconut palm were used in this study. All the tested Ganoderma isolates were found to grow faster on Malt Extract Agar MEA amended with oil palm extract and oil palm extract media compared to MEA coconut extract and coconut extract with MEA. Among seven Ganoderma isolates G. lucidum was recorded to have the lowest growth rate. Brownish discolouration zone outside the mycelia growing point was noted for all the Ganoderma isolates on coconut extract and MEA supplemented with coconut extract media. In another study sugar compounds for both oil palm and coconut extracts were analyzed using HPLG. Oil palm;s extract sugar profile: 1.7 glucose and 0.72 fructose. Coconut;s extract sugar profile: 2.54 glucose 17.3 sucrose and 1.29 fructose. Media with three and seven different combinations of sugar were prepared based on the sugar profiles obtained from oil palm and coconut extracts. Most of the Ganoderma isolates especially G. boninense from oil palm Ganoderma species from coconut palm and G. australe from Schizolobium parahybum showed better mycelia growth on media with higher level of sucrose fructose or both sucrose and fructose. On the other hand G. lucidum was found to have better growth on the media with glucose or in combinations with glucose. Further studies are required to identify the optimum amount of respective sugar compounds to produce optimal growth in all Ganoderma isolates.
