Anti-inflammatory activity of four Cryptocarya species Lauraceae

Abstract

Cryptocarya genus with 480 species belongs to Lauraceae family. Some have been used as traditional medicine to relieve sore muscles and headache fever diarrhea postpartum care anti-fungal and anti-bacterial. Presence of secondary metabolites such as flavonoids pyrones Iignans terpenoids alkaloids and steroids are reported with 2-pyrones and flavonoids being the most common. The anti-inflammatory properties of Malaysian Cryptocarya species has not yet been reported thus the lipoxygenase LOX xanthine oxidase XOD and hyaluronidase HYA inhibition assays were performed on four Cryptocarya species namely C. ferrea var ferrea C. niara C. obliqua and C. rugulosa. The final concentration of 100 g/ml was used for samples in all three assays. All reactions were performed in triplicate in 96-well micro-plate in Tecan Infinite M200 Microplate Reader Tecan Austria. The IC‚‚ values were then calculated using the Probit Analysis program. C. ferrea var ferrea extracts exhibited the highest hyaluronidase inhibitory activities with 100 inhibition while the other three demonstrated low inhibitions. C. nigra showed high inhibition of lipoxygenase enzyme activity at 86 C. obligua was inactive. Additionally the root methanol extracts of C. ferrea var ferrea and C. rugulosa showed moderate enzyme activity at 64.06 and 54.62 inhibition of Iipoxygenase respectively. The C. ferrea var ferrea and C. nigra extracts exhibited moderate xanthine oxidase inhibitory activity with 65.97 and 66.59 inhibition respectively while C. rugulosa displayed low inhibition at 36.46 . Results from this anti-inflammatory evaluation suggests that C. ferrea var ferrea and C. nigra may have great potential for further development as therapeutic agents or food supplements.
