Abstract
Intracellular lipase is naturally immobilised to the mycelial membrane of Aspergillus niger. Studies were made on various parameters: incubation time weight of mycelia and pH in relation to lipase activity. About 60 of the initial activity was retained after 6 days of continuous reaction. The mycelia were also entrapped within calcium alginate gel. 60 activity was retained on immobilisation and it was found that 40 was retained after 17 days of continuous reaction. Crude extracellular lipase was immobilised by adsorption onto various matrices such as cellulose DEAE - and CM-Sephadex Celite Kieselgur G and controlledpore glass CPG at various pH. Activity was retained but stability was low in all cases. Covalent attachment of the enzyme was done on DEAE-Sephadex but activity retained was low.d
