Abstract

Crude lipase was purified 11 fold with 40 percent recovery in a four-step conventional process using water extraction ammonium sulphate precipitation and fractionation through Sephadex G-25 and G-200. Using the FPLC system the crude lipase was purified in a single step using Superose 6 Mono-Q HR5/5 or Mono-P HR5/5 columns. With Superose 6 a 12-fold purification and about 90 percent recovery was obtained. With Mono-Q and Mono-P columns the recovery and purification folds were lower but in both cases two active lipase peaks were obtained.
