Abstract
The effects of tentacle extract of the jellyfish Catostylus mosaicus on nerve-muscle was studied on rat phrenic hemidiaphragm and chick biventer cervicis preparations. The crude tentacle extract caused an irreversible 70 percent depression of the twitch amplitude of the phrenic nerve-diaphragm preparation that was indirectly stimulated. Treatment of tentacle extract with trypsin caused complete inhibition of muscle contraction when the hemidiaphragm was stimulated indirectly. This inhibition was also irreversible and shown to be myotoxic as the muscle did not respond to subsequent direct stimulation. Further evidence of the myotoxicity of the tentacular extract was the observation that it caused a 75 percent reduction in the twitch amplitude of this preparation elicited by direct stimulation. The tentacle extract caused a complete and irreversible inhibition of the twitch response of the chick biventer cervicis preparation accompanied with a delayed muscle contracture which suggests that the tentacle extract caused depolarization of the muscle. However the addition of exogenous acetylcholine produced a twitch response to the tentacle extract treated muscle indicating that the inhibition caused by the latter is presynaptic therefore neurotoxic.
