Abstract

Sporulation of Oncobasidium theobromae the causal agent of vascular streak dieback of cocoa was induced in culture for the first time recently. The fungus was grown first on Corticium culture medium CCM and then on 2 percent water agar contained in a dual compartmented petri dish. The cultures were incubated at 25 aprroximately 2 degree celsius and aerated continuously with saturated moist air. Powdery-white aerial mycelial aggregations appeared on the surface of the 2 percent water agar within 8-10 days of incubation. Examination under a light microscope revealed basidial proliferations elongation of epibasidial sterigmata and basidiospore formation within the mycelial aggregationns. Further detail studies on the basidial and basidiospores morphology were carried out with the aid of a scanning electron microscope. The hymenium remained fertile for 4-6 days with at least three rounds of fruiting. Later observations showed that basidiospore production could also be induced in culture by transfering mycelial discs of 10 or 14 mm from advancing margin of the colony growing on CCM to Petri dishes containing 2 percent water agar and incubating the plates under previuosly described conditions. Basidiospores formation occurred within 4-5 days of incubation
