Abstract
Using two methods 2 barches of paraffin oil named old and new paraffin oil were tested for toxicity before they were used as oil overlays in embryo culture studies. In the first method epididymal sperm were collected from mature male mice and were placed in microdrops of culture medium which were covered with the respective oil overlays. Using an inverted microscope observation was made to check the survival of the sperm under study. In the second method eight-cell stage mouse embryos were cultured in vitro using the respective oil overlays to be tested. A series of time-lapse photomicrographs was taken at 0 0.5 1 24 and 48 hours in culture. In the sperm motility assay the results obtained from the old batch of paraffin oil showed that the sperm became rapidly inactive and were dead within 10 minutes while those cultured in the new batch of oil survived until the next day. In the embryo culture study all the embryos cultured in the old paraffin oil started to show a pronounced blastomere swelling effect and disintegrated after half an hour in culture. At 1 h in culture the blastomeres appeared to have lysed. In contrast embryos cultured in the new batch of paraffin oil developed to the blastocyst stage. In conclusion care must taken in choosing an oil overlay during embryo culture in vitro especially the paraffin oil to ensure that it is not toxic to the embryos in culture
