Abstract

Techniques to develop PAL gene expression in the cells and tissues of Cucumis melo L. were established using particle gun electroporation technique. These techniques enable us to study the regulation of PAL promoters isolated from pea Pisum sativum in a heterologous plant system; pPaL1 and pPaL2 were shown to be expressible in melon. The effect of UV radiation on the regulation of the PAL promoters under different regime of UV irradiation was tested on the mesophyll protoplasts of melon. Maximum induction of GUS expression was observed in PSPAL1 and PSPAL2 at 2 min and 80 sec respectively. Further confirmation of the GUS expression in the tissue of melon was carried out using particle. Similar pattern of induction of GUS expression by UV radiation has been regulated by the PSPAL1 and PSPAL2 promoter. Thus this suggests that similar mechanism of PAL gene expression may be present in plant
