Abstract
Pycnospore germination fluid of Mycosphaerella pinodes a fungus pathogenic on pea contains both elicitor and suppressor for the accumulation of pisatin a major phytoalexin of pea. Concominant presence of suppressor with elicitor results in delaying the accumulation of PAL the gene encoding phenylalanine ammonia-lyase-mRNA and CHS the gene encoding chalcone synthase-mRNA in pea epicotyl tissues. Nuclear run-on assay revealed that M. pinodes-suppressor and orthovanadate an inhibitor for ATPase temporally depressed the transcriptional activation of PAL and CHS genes induced by elicitor in pea epicotyl tissues. To characterise nuclear factors involved in activation and deactivation of plant defence genes by signal molecules secreted from a fungal pathogen. TATA-proximal region of the promoter of phenylalanine ammonia-lyase gene 1 PSPAL1 and 2 PSPAL2 and chalcone synthase gene 1 PSCHS1 and 2 PSCHS2 were dissected and cis-regulatory elements were examined by gel mobility-shift assays and in vitro DNase I-footprinting analysis. Changes of chromatin structures were found at multiple sites involved in elicitor-mediated activation. Among them prominent changes were seen at a region containing consensus sequence motifs of box 1 G-box and box 2 for elicitor-mediated activation and the binding of elicitor-inducible nuclear factors was competed with the fragments from other elicitor-inducible promoters
