Abstract

Thioesterase showing relatively high activity towards palmitoyl-ACP has been implicated in palmitic acid accumulation in the oil palm mesocarp. Nevertheless the characterization and regulation of this enzyme need to be investigated to understand the possible involvement of the thioesterase in this experiment. Thus present study involve the isolation and characterization of this enzyme. Acyl-ACP thioesterase was extracted from oil palm mesocarp and partially purified by ammonium sulphate precipitation anion exchange chromatography on DEAE-HR and hydroxyapatite chromatography column. The thioesterase was purified ten fold with specific activity of 2563.11 U/mg. The enzymes showed activity towards palmitoyl-ACP and oleoyl-ACP with negligible activity towards stearoyl-ACP. The enzymes also showed activity towards acyl-CoA substrates
