Abstract
Lectin from Artocarpus integer Merr. seeds has been purified using rivanol 6 9-diamino-2-ethoxyacridine lactate and the yield was 1.23. The polyacrylamide gel electrophoresis PAGE gave a long band of estimated molecular weight of 103 500. Sodium dodecyl sulfate-PAGE SDS-PAGE gave three components of 24 500 30 00 and 75 800. The lectin consists of four polypeptide chains composed of two light beta- chains 24 500 and two heavy Beta-chains 30 000 of the alpha-2 Beta-2 form. Artocarpus integer lectin possesses the sugar binding site specific for alpha-D-galactosyl moiety since its hemagglutinating activity was inhibited by the simple sugar alpha-D-galactose/N-acetyl-alpha-D-galactosamine and its glycoside derivatives. Of the sugars tested the lectin showed the strongest affinity for methyl alpha-D-galactopyranoside followed by methl 3-0-alpha-D-galactopyranosyl-alpha-D-galactopyranoside
