Pyrophosphatase activity in the latex of Hevea brasiliensis

Abstract

A non-specific acid pyrophosphatase was isolated from the latex of Hevea brasiliensis. This enzyme possessed an isoelectric point of 4.50 did not depend on Mg2 for its activity and was active between pH 4.2 - 6.2 with an apparent pH optimum of 5.0 when inorganic pyrophosphate was used as the substrate. This enzyme was capable of hydrolysing several phosphorylated compounds. Subcellular fractionation of fresh latex indicated that this enzyme was mainly located in the B serum suggesting that it was of lutoidal origin. This enzyme exibited remarkable stability at 4 deg. C between pH 4 -7. The molecular weight of this enzyme was estimated by gel filtration chromatography to be approximately 60 000. The enzyme was insensitive to the thiol reagent p-chloromercuribenzoate
