Abstract

Calmodulin was isolated from the hepatopancreas of Achatina fulica by phenyl-Sepharose affinity column chromatography. Calmodulin accounted for 1.2 of the total protein in the heat-treated extract. It has a molecular weight of 16 800 and pI value of 3.9. Mobility studies revealed that the protein migrated faster in the presence of EGTA than it did in the presence of calcium ions under non-denaturing electrophoretic conditions. Absorption spectral scan of the purified protein showed peaks at 259 265 268 and 278 nm. Upon incubation of the purified calmodulin with the ovatesticular microsomal fraction in the presence of various factors such as calcium EGTA and trifluoperazine it was evident that calmodulin in the presence of calcium ions increased the production of testosterone by almost 2.5-fold but had little or no effect on the production of other steroids
