Abstract

Capsicum annuum L. seeds were aseptically germinated on a half strength of Murashige and Skoog salts Flow Lab. Australia medium containing 30g/L sucrose and 2.5g/L Gelrite agar pH 5.8 26 deg. C in dim light. Explants from the chilli seedlings e.g. hypocotyl leaf and root were cultured on a modified MS salts agar medium Dix and Street 1976 containing 0.4 mg/L 2 4-dichlorophenoxy acetic acid 2 4-D; 0.03 mg/L kinetin KN 30 g/L sucrose and 2.5 g/L Gelrite agar; pH medium 5.8 in order to initiate and establish callus formation. The present experiments show that after a week of the culture the hypocotyl explants induced more callus formation than the other parts of the seedlings. The hypocotyl segments induced 60 of callus formation while the leaf segments induced only 10 callus after one week culture. After 2 weeks of culture the induction of callus formation for the hypocotyl leaf and roots was increased to 90 80 and 40 respectively. The callus is then regularly maintained on the modified MS agar medium. The effects of Mancozeb as Manzate powder contained 80 Mancozeb on chilli hypocotyl growth on modified MS media containing various concentrations of the fungicide were studied. The callus growth and development of the chilli tissue was investigated. The callus was also biochemically assessed at weekly basis until 4 weeks for the production of soluble protein and ascorbic acid
