Abstract

Calli can be obtained from the embryos and young fruits and the stems and leave petioles of in vitro-grown plantlets of Murraya koenigii. The embryos and fruits were cultured on the Linsmaier and Skoogs LS medium containing 2.0 mg/L 2 4-dichloro-phenoxy acetic acid 2 4-D 1 polyvinyl-pyrolidone PVP while calli from the stems were formed on the same medium but supplemented with 3.0 mg/L 2 4-D 1 PVP or with 2.0 mg/L benzyl amino purine BAP or with 3.0 mg/L napthalene acetic acid NAA. Induction of calli from the petioles required the presence of 4.0 mg/L 2 4-D 1 PVP. Sub-culture of calli was best carried out on the same medium used for a particular explant containing either PVP or charcoal and placed under light or in the dark. The capillary GC profile of compounds extracted by Simultaneous Distillation Extraction SDE technique from the fruit callus was different from those obtained using the leaves and fruits of plant
