Abstract

Until the advent of high-throughput genomic technologies molecular analysis of Hevea genes of interest was conducted on a gene-by-gene cloning approach. The generation and analysis of latex ESTs has been performed by the RRIM since the year 2000. Over the years. this venture was greatly facilitated through collaborations between researchers from the RRIM the Malaysia Genome Institute the Korea Kumho Petrochemical Co. Ltd. and Universiti Putra Malaysia including funding from the Malaysian Rubber Board and the Malaysian Ministry of Science Technology and Innovation. The objective of this project is to create a vast molecular resource which would serve as a pool of expressed sequences for various aspects of rubber research thus circumventing multiple cloning experiments. In addition annotation of ESTs in this molecular resource allows it to be a tool for the discovery of novel genes. To date the RRIM has generated more than 20 000 ESTs from a latex cDNA library from the tree clone RRIM 600. Currently this is the largest latex EST collection not just in Malaysia but also within the global rubber research community. Analysis of latex ESTs has provided a profile of genes that are expressed in the laticifer and subsequently selected ESTs were applied in studies on rubber biosynthesis and latex allergy. In 2005 latex ESTs were compiled into NRESTdb Natural Rubber EST Database; http://genome.ukm.my/nrestdb/ an online genomic resource for scientists working on biotechnology of the rubber tree. NRESTdb resources include EST sequences and assemblies curated databases based on KOG and GO functional classifications and search tools to facilitate user navigation. NREST db has also been registered in the Molecular Biology Database Collection of the Nucleic Acids Research Journal since January 2007. In future NRESTdb resources will be continually upgraded in line with the parallel expansion of the RRIM EST collect ion and according to user needs.
