Abstract

CB Crossbred G4 horse is result of crossing between the Indonesian sandel female and thoroughbred male horse that already get four generation by PORDASI The Indonesian . Equistrian Association has categorized as Indonesian horse with level body style and running capability similar with thoroughbred. CG Chorionic Gonadotropin from horse that is eCG is a hormonal preparation that is quite effective to treat ovarian hypofunction. in cows. Until now raw material for eCG production available on the market still from thoroughbred horse serum 3-4 months pregnant. To improve the biologist capability of eCG in eluate fraction of sephadex G-75 separated 3.5 months pregnant Indonesian CBG4 mares serum be needed to minimize the contain of other honnone E2 for increasing degree of eCG purification. The research has done with aim to measure the content of E2 Estradiol-17 Beta measurement with RIA technique in the eluate number one until eleven separation product of CBG4 3.5 months pregnant by sephadex G-75. The result has proven that the sequence of eluate fraction of separation CBG4 3.5 months pregnant by sephadex G-75 give a significant support p 0.01 to E2 content measured by RIA technique. This fact describe that the higher sequence of eluate give the higher E2 content. In this research the last eluate sequence or fraction number eleven has the highest 372.00186.31 pg/ml E2 content and the eluate sequence number three has the lowest 9.17 12.00 pg/ml E2 content. Practically the volume of eluent flowing in to coloumb can be increased and make taller of sephadex G-75 coloumb. Both effort can minimize canalization effect increase the number of eluate sequence stored and give higher chance to the whole component in eluent for completely mixed appropriate with each molecule size.
