Antibody profile of Indonesian strain vaccine and neutralization test against sub-cluster H5N1 virus

Abstract

The pandemic threat posed by highly pathogenic H5N1 influenza A virus has created an urgent need for vaccines to protect against H5N1 virus infection. Indonesia has been especially affected. More than 140 cases of human infection and mortality rate are the highest reported to date accounting for more than a third of all confirmed human H5N1 infections worldwide. The purpose of this research was to analyze antibody profile and the effectiveness of the seed human vaccine to the animal experiment. We vaccinated three kinds of animals in Animal BSL-3 Airlangga University mice guinea pig and non human primates Macaca fascicularis with Indonesian H5N1 Subtype Vaccine. Twenty days after vaccination the animals given a booster vaccination with the same seed vaccine to get maximum antibody response. Antibody profile was analysed by HI test and Neutralization test. Results of this research showed that Indonesian H5N1 Subtype Vaccine induced antibody in the animals with different titers and was able to neutralize virus A/lnd/TDC/05 H5N1 and A/Ck/ Ind/114/08 H5N1. The challenge test with H5N1 subtype avian influenza virus from humans and animals is still in progress.
