Development of lymphomas in male sprague dawley rats exposed to N-methyl-N-nitrosourea

Abstract

The aim of this study was to establish a lymphoma model in Sprague Dawley rats. A total of thirty two 8-week-old male Sprague Dawley rats were allocated into two groups the control group and the MNU group n 16. The MNU group was injected intraperitoneally with N-methyl-N-nitrosourea MNU at a dose of 60 mg/kg of body weight twice weekly for two consecutive weeks. The rats were sacrificed at week 20 of the experimental period. On necropsy lymph nodes thymus spleen liver kidneys lung heart and intestines were examined for evidence of lymphoma. Results showed 4 of 16 rats induced with MNU had lymphadenopathy splenomegaly and hepatomegaly. One of the rats had enlarged kidneys. Microscopic examination of the necropsied organs revealed massive proliferation and infiltration of neoplastic lymphocytes in the lymph nodes spleen liver kidneys lung and heart. Results were consistent with lymphoma. In conclusion a lymphoma model was successfully established in male Sprague Dawley rats exposed to MNU.
