N-Methyl-N-Nitrosourea induced leukaemia in male Sprague Dawley rats

Abstract

N-Methyl-N-nitrosourea MNU has broad spectrum of target organs including Iymphohaematopoeitic system. Several studies have shown MNU induced leukaemia in F344 rats. The study was conducted to investigate the incidence of leukaemia in 16 young adult male Sprague Dawley rats administrated MNU intraperitoneally i.p at a dose of 60 mg/ kg of body weight twice weekly for two consecutive weeks. The rats were kept for 20 weeks before blood samples were taken and examined for the presence of leukaemic cells. Results showed 2 of 16 rats died at week 16 of the experimental period and 4 of 14 rats had marked lymphocytic leukocytosis. The rats with marked lymphocytosis were also severely anaemic and blood smear examination revealed numerous lymphocytes and lymphobasts with prominent criteria of malignancy which was consistent with leukaemia. Blood smear of rats with slight lymphocytosis contained predominantly atypical lymphocytes. In conclusion administration of MNU to male Sprague Dawley rats induced leukaemia at a relatively low prevalence rate at 5 months post-exposure.
