Abstract
An extract derived from a combination of the herbs Tinospora crispa Momordica charantia Alpinia galanga and Portulaca quadrifida has been claimed to be an effective remedy for diabetes mellitus. The objective of this study was to verify the efficacy of the extract as an anti-diabetic agent. The freese-dried aqueous extract was tested on normal rats in an Oral Glucose Tolerance Test OGTT. In a range of dosages comprising 2.5 25 250 500 and 2500 mg/kg b.w. potential hypoglycaemic effect was apparent at the higher dose of 500 mg/kg b.w. Although 14 per cent of the extract consisted of glucose doses of 500 and 2500 mg/kg b.w. caused lower average rates of increase 1.03 plus minus 0.25 and 0.69 plus minus 0.21 mg per cent/min respectively compared to saline 1.72 plus minus 0.49 mg per cent/min. Their peak glucose concentrations were not significantly different from saline and the AUC values from peak to 210 minutes were also lower 20780.36 plus minus 1983.14 and 22985.82 plus minus 1636.69 mg per cent/min respectively compared to saline 25611 plus minus 1284.94 mg per cent/min. The hypoglycaemic activity of the extract could be due to the enhancement of beta-cell sensitivity to the insulin-releasing effect of glocose the improvement of insulin action and the extra pancreatic effects of glocose-6-phosphate dehydrogenase activation or suppression of hepatic glucose production.
