Abstract

Callus tissue culture of Citrus hystrix DC.Rutaceae was aseptically initiated from stem explant. The formation of callus derived from explants could be optimised by manipulating the plant growth regulators in a basal medium of Murashige and Skoog MS. A series of MS medium was set up with a combination of 0.20 mg/l 2 4-dichlorophenoxy acetic acid and 1.50 mg/l benzylaminopurine acid in the dark and 0.50 mg/l 2 4-dicholophenoxy acetic acid and 1.00 mg/l benzylaminopurine acid in the light. The callus formed was friable and yellowish in colour. Optimal callus growth was observed during the fourth week under dark condition and third week under light condition. Natural flavonoids hesperitin and quercetin were found in the callus tissue culture of C. hystrix.
