Abstract

DNA extraction protocols with minor modification have been used to get high purity of genomic DNA and reproducible results. Two types of PCR-based DNA markers LP-RAPD Long Primer-Randomly Amplify Polymorphic DNA and AFLP Amplified Fragment Length Polymorphism were applied to identify and characterize 12 accessions of C. asiatica. In LP-RAPD study optimum concentration and conditions for the protocal have been obtained. Amplification using long primers that each comprosing of 18 to 24 bases will generates 6 to 30 DNA fingerprints. Six out of ten single primers ERICIRERIC2 PehA3 PehA6 Puc/M13F BOXIR were selected for further LP-RADP analysis. In AFLP study the screening of 64 primer combinations on CA3 and CA6 are still in the progress.
