Abstract
Plantlets were obtained by culture of orchid protoplasts. Callus of Phalaenopsis Wedding March was pre-cultured on sucrose-free medium for 3 weeks. Protoplasts were isolated from the pre-cultured callus. About 10 of the protoplasts cultured in sorbitol or maltose containing media divided within 4 weeks. Eleven months were required for Phalaenopsis protoplasts to produce PLB about 2-3 mm in size. About 100 PLBs were obtained from 1.5 x 10 protoplasts. These PLB when transplanted onto Kyoto medium developed into plantlets which were transplanted and successfully grown under greenhouse conditions.
