Abstract

Germplasm conservation of species producing recalcitrant seeds is a problem and poses a great challenge. Despite cryopreservation being mentioned as the only possible method for conservation of such materials success has been limited and the sporadic survival is often not reproducible. The approach to cryopreservation is usually through empiric means by screening for tolerance to various cryoprotectant cocktails and testing for the best exposure duration. This results in little understanding of the effect of the substance used to the embryos. This study details the effect of exposure to vitrification cocktail prior and subsequent to liquid nitrogen exposure on the structural changes in rambutan zygotic embryonic axes as compared to the structure of freshly excised embryonic axes. Severe damage to cell structure was observed when freshly excised embryonic axes were cryopreserved without pretreatment. The pretreatment with vitrification cocktail helped to reduce damages to overall cell or tissue structure of the embryonic axis after cryopreservation.
