Abstract

An experiment was carried out to determine the most suitable concentration of BAP 6-Benzylaminopurine and NAA Napthaleneacetic Acid for direct shoot regeneration from petiole and leaf explants of papaya cv. Eksotika. Explants were cultured on MS Murashige and Skoog medium supplemented with 500mgL-1 caseine hydrolysate 0.2 phytagel and different combinations and concentrations of BAP and NAA. For petiole explant MS medium containing 1.0 mgL-1 BAP and 0.05 mgL-1 NAA produced the highest percentage of shoot formation 60.24 and mean number of shoots per explant 9.5 while MS medium containing 0.1 mgL-1 BAP and 0.05 mgL-1 NAA resulted in the highest shoot length 0.69 cm. No shoot formation occurred on MS medium without BAP. With leaf explants MS medium containing 1.0 gL-1 BAP and 0.05 mgL-1 NAA produced the highest percentage of shoot formation 70.07 and mean number of shoots per explant 46.5. The treatment also showed significant difference on mean number of shoots formed per explant compared to other treatments including the control. The treatment also resulted in high shoot length 0.48 cm. Shoot formation did not occur on MS medium without BAP.
