Abstract

Isoflavones which are present in soybean and its fermented product (tempe) are known as having oestrogenic and antioxidant properties. Thin Layer Chromatography (TLC) was applied to the isolation and purification of isoflavones. The packing material of silica-gel and mobile phase of ethyl acetate-formic acid-water (10:2:3) were used for isolation while purification was in a mixture of chloroform-methanol (9:1). Quantification of the components obtained was carried out spectrophotometrically. Soybean fermented with R. oryzae contained 4.9 mg genistein 3.8 mg daidzein. 19.9 mg genistin and 21.6 mg daidzin per 100 g. When R. oligosporus was used as inoculum the amounts of genistein daidzein genistin and diadzin were 13.8 mg 12 9 mg 10.0 mg and 8.1 respectively. The concentration of isoflavones in unfermented soybeans was also determined for comparison
