Abstract

Proteolytic activities in the cotyledons of winged bean (Psophocarpus tetragonolobus (L.) DC.) after 7 days of germination were studied at different pHs using casein as a substrate. Results showed that there are two pH optima for protease activity at pH 5.0 and pH 6.5 inferring that there are at least two types of proteases in winged bean cotyledons. During germination the activities at both pHs increased after 2 hours of soaking and started to decrease after 24 hours. During the early part of germination up to the 4th day the activity of pH 6.5 protease is generally lower than that of pH 5.0. After this stage the activity of pH 5.0 decreased to below that of pH 6.5 and increased again after the 6th day then stabilised after the 8th. pH 6.5 protease on the other hand increased after day 4 to a level higher than pH 5.0 then decreased again after day 6.
