Abstract

The production of lipase from Corynebacterium sp. was carried out using an optimized medium containing (percent w/v): crude palm oil 0.6; sucrose 0.4; Tween 20 1.0: yeast extract 1.5; and various mineral salts. the pH of the medium was adjusted to pH 4.0 cultivation temperature was at 37 degrees C and inoculum size of 5 percent (v/v). An increase in enzyme activity of about 80 percent was obtained after optimization was performed. Lipase production in the fermenter system (agitation rate: 250 rpm; aeration rate: 1 vvm) by Corynebacterium sp. under the optimized conditions was 2.17 umol free fatty acid ml-1 min-1. This was an increase of about 197 percent enzyme activity compared to the shaking flask culture system prior to optimization. Hydrolysis with lipase from Corynebacterium sp. was optimum at pH 7.0 and temperature 50 degrees C. The enzyme activity was also enchanced by the presence of calcium ions and sodium taurocholate.
